Das vorliegende Forschungsprojekt wurde mit Ratten als Versuchstiere durchgeführt. Die Studie dauerte sechs Monate und umfasste die orale Verabreichung des Verhütungsmittels LD, wobei jede S.C.-Tablette 0,15 mg Levonorgestrel und 0,03 mg Ethinylestradiol enthielt. Die Studie wurde an zwei Gruppen von acht männlichen und acht weiblichen Ratten durchgeführt. Die erste Gruppe diente als unbehandelte Kontrollgruppe, die zweite Gruppe wurde behandelt. In den ersten drei Monaten erhielten die behandelten Ratten oral eine in Wasser aufgelöste Tablette, in den letzten drei Monaten wurde die Dosis auf eine Tablette pro Tier erhöht. Die histopathologischen Veränderungen bestanden in einer Unterdrückung der Spermatogenese in den Hoden bei den Männchen und einer Atrophie der Eierstöcke bei den Weibchen. Darüber hinaus gab es histopathologische Veränderungen in Leber, Niere, Herz, Lunge, Skelettmuskulatur, Haut, Nebenhoden und Prostata.
